Clinical Manifestations in Children with Staphylococcal Bacteremia Positive for Panton-Valentine Leucocidin: A Nationwide Survey in a Low Methicillin-Resistance Setting.

Allan Bybeck Nielsen, Anders Rhod Larsen, Andreas Petersen, Henrik Carl Schønheyder, Henrik Westh, Thomas Benfield, Ulrikka Nygaard,

A total of 714 pediatric cases of Staphylococcus aureus bacteremia were identified from 2008 to 2015 in Denmark; 98% were methicillin-susceptible S. aureus (MSSA). Fifteen isolates (2,1%) were Panton-Valentine leucocidin positive (0.17/100,000 children/year) and 87% MSSA. Eight cases (53%) were severe, including all pneumonia cases. Panton-Valentine leucocidin positive Staphylococcus aureus ... Read more >>

Borderline-Positive Quantiferon in Children.

Ulrikka Nygaard, Anja Poulsen, Alex Yde Nielsen, Pernille Ravn, Nadja Hawwa Vissing,

We systematically retested children with borderline-positive quantiferon (0.35-0.99 IU/mL) during the period 2015-2019. Among 647 tests, 27 (4%) were positive (>0.35 IU/mL). Borderline-positive quantiferon accounted for 10 of 27 (37%) positive tests. When retested, 9 of 10 (90%) were negative. Children younger than 5 years had negative tuberculin skin tests. ... Antibody response to vaccination after haematopoietic cell transplantation in children using a reduced dose schedule-A retrospective cohort study.

Lotte Jensen, Anja Poulsen, Ulrikka Nygaard, Thomas Scheike, Margit Schilling Riis, Freddy Karup Pedersen, Carsten Heilmann,

Children receiving HCT loose protective immunity to vaccines received pre-HCT. Therefore, revaccination post-HCT is of major importance. In Denmark, a vaccination schedule with fewer doses post-HCT has been used, including two doses for diphtheria, tetanus, polio, measles, mumps, and rubella, and one dose only for Haemophilus influenzae type B. The standard practice in paediatric departments in Danish hospitals is to prescribe hypotonic maintenance fluids (sodium content 20-40 mmol/l) for children, who are fasting or have a reduced enteral intake. The past decades have provided strong evidence, that this can lead to hypo-natraemia and subsequent neurologic damage or death. We ... Read more >>

Hepatotoxicity is a known toxicity to treatment of childhood acute lymphoblastic leukemia. Hepatotoxicity occurs during maintenance therapy and is caused by metabolites of 6-Mercaptopurine (6 MP) and Methotrexate (MTX). Our objective was to investigate the association between alanine aminotransferases (ALAT) levels and relapse rate. We included 385 patients enrolled in ... Read more >>

Myelotoxicity after high-dose methotrexate in childhood acute leukemia is influenced by 6-mercaptopurine dosing but not by intermediate thiopurine methyltransferase activity.

Purpose
Through enhancement of 6-mercaptopurine (6MP) bioavailability and inhibition of purine de novo synthesis, high-dose methotrexate (HD-MTX) may increase incorporation into DNA of 6-thioguanine nucleotides, the cytotoxic metabolites of 6MP. Patients with intermediate activity of thiopurine methyltransferase (TPMT(IA)) have higher cytosol 6-thioguanine nucleotide levels. We investigated toxicity following HD-MTX during MTX/6MP ...
